This application note presents how to perform cell culture on chip using the Cell and Biology Pack and the MicroSlides developed by ALine Inc.
This systems uses continuous perfusion, creating a stable environment for the cells, without any manual operations. Inject up to 10 different liquids. Program injection sequences and automate your experiments for better reproducibility. Suitable for 3D cell culture and dug screening.
The MicroSlides are designed to avoid applying shear stress on your cells. The cells are not directly put into the flow.

A bubble trap can be added just before the MicroSlide to make sure that air bubbles don’t enter the chip.
It is also possible to perform a medium switch in order to expose the cells to different drugs or conditions. Check out our dedicated application note for this specific setup.
Before starting the experiment, rinse the MicroSlide, the reservoirs and all the tubes and connectors with 70% ethanol to ensure sterility. Please make sure to perform all the following steps under a biosafety hood to avoid contamination.
Fill the reservoir with the culture medium and connect the reservoir to the flow controller.
Connect the reservoir to the MicroSlide.
Once the MicroSlide is connected to the Perfusion Pack, tilt the device as shown on the picture. Using the Elveflow Smart Interface Software, activate the pump until all three wells are ¼ filled then shut off.
Add 10- to 30 uL of sample to each well with a micropipette.
Remove the adhesive liner from the MicroSlide, and seal it with the lid. Use thumb pressure to seal the lid.
During the experiments, the MicroSlide and the reservoirs can be placed inside an incubator or environmental chamber, while the OB1 and flow sensor stay outside. Use longer pieces of tubing in order to place the instruments outside the incubator.

Fluorescence reader for microfluidic qPCR: faster, more sensitive and less expensive than most optical microscopes, it is a smart alternative for real-time fluorescence measurements of your on-chip qPCR signal.
Prostate cancer is the second leading cause of cancer-related death for men. Circulating tumor cells (CTCs) are considered as a marker of early cancer diagnosis and disease severity. Their screening in blood is thus crucial to detect metastatic stage in cancer patients.
Until recently, microfluidic devices have been employed to support tissue-engineering experiments on basal lamina, vascular tissue, liver, bone, cartilage and neurons as well as organ-on-chips.
